Investigations on Gelatin. Part VIII. The Osmotic Pressure of Gelatin in Solutions of Sodium Salicylate.

چکیده
THE present investigation was undertaken under the direction of Professor S. B. Schryver with a view to ascertaining the osmotic pressure of gelatin. Previous workers on this problem have not obtained their gelatin absolutely free from ash. Although the view is now commonly held that a low ash has no direct influence on the osmotic pressure it was thought desirable, since gelatin of exceptional purity has recently been prepared in this department [1923] to study the osmotic pressure of this preparation. Since the osmotic pressure of gelatin is extremely low it is necessary to employ a fairly strong solution in order to obtain a pressure which can be easily and accurately measured with a very simple osmometer such as has been used in this investigation. If water be used as the solvent it is not possible to work at ordinary room temperature owing to the formation of a gel. It was therefore decided to employ as the solvent of the gelatin a solution of sodium salicylate, which, whilst having no chemical action upon the colloid, prevents gel formation. The liquefaction of gelatin by certain salts has been proved to be entirely reversible by Briggs and Hieben [1920] and the present investigator finds this to be the case with sodium salicylate. This reversibility proves that the salt in question has no chemical action upon the gelatin. The presence of this salt also serves to keep the solution sterile. A strong solution of sodium salicylate has therefore been used as the solvent of the gelatin in all the experiments herein described.

The Isoelectric Point of Gelatin at 40°c
Measurements have been made at 40 degrees C. of the osmotic pressure and viscosity of 1 per cent gelatin solutions containing varying amounts of hydrochloric acid or sodium hydroxide. Each property was found to exhibit a decided minimum near pH 4.7. In the osmotic pressure experiments the pH of the inside solutions was greater than that of the outside solutions at pH values below 4.7, while it ...
متن کاملThe Swelling and Osmotic Pressure of Gelatin in Salt Solutions
1. The swelling and the osmotic pressure of gelatin at pH 4.7 have been measured in the presence of a number of salts. 2. The effect of the salts on the swelling is closely paralleled by the effect on the osmotic pressure, and the bulk modulus of the gelatin particles calculated from these figures is constant up to an increase in volume of about 800 per cent. As soon as any of the salts increas...
متن کاملThe Mechanism by Which Trivalent and Tetravalent Ions Produce an Electrical Charge on Isoelectric Protein
1. Experiments on anomalous osmosis suggested that salts with trivalent cations, e.g. LaCl(3), caused isoelectric gelatin to be positively charged, and salts with tetravalent anions, e.g. Na(4)Fe(CN)(6), caused isoelectric gelatin to be negatively charged. In this paper direct measurements of the P.D. between gels of isoelectric gelatin and an aqueous solution as well as between solutions of is...

1. It is shown that when part of the gelatin in a solution of gelatin chloride is replaced by particles of powdered gelatin (without change of pH) the membrane potential of the solution is influenced comparatively little. 2. A measurement of the hydrogen ion concentration of the gelatin chloride solution and the outside aqueous solution with which the gelatin solution is in osmotic equilibrium,...
